Our earlier studies suggested that human monocytes, stimulated with lipopolysaccharide (LPS), may be important intermediaries in autologous B cell transformation. Subsequently, however, we failed to detect significant amounts of Interleukin-1 in the LPS-monocyte coculture and in other experiments, we were unable to demonstrate expression of Interleukin-1 in monocytes by immunocytochemistry. To clarify our observations and to examine IL-6, a monocyte derived cytokine that is involved in B cell activation, here we examined IL-6 levels in supernatants of LPS-stimulated monocyte-B cell cocultures, and in monocyte/lymphoid cultures stimulated with LPS alone. We found that the monocyte/B cell cocultures and LPS-stimulated monocyte cultures consistently produced large amounts of IL-6. Transformed lymphoid cells were not capable of producing IL-6. Furthermore, the addition of mitogen-stimulated autologous lymphoblasts to LPS-stimulated monocytes did not result in IL-6 production. These observations suggest that human monocytes stimulated with LPS are capable of producing IL-6, and, therefore, may be an important intermediary in the autologous transformation of human B cells.?
